Effects of Seed of Cart ham us tinctorius L. on Gingival fibroblasts, Periodontal ligament cells and Osteoblasts


Abstract
The purpose of this study was to evaluate the effects of Carthamus tinctorius L on gingival fibroblasts, periodontal ligament cells and osteoblasts. Periodontal ligament cells and gingival fibroblasts were primarily from extracted premolar with non-periodontal diseases. 0steoblasts were getten from calvariae of rat. Cells were cultured with DMEM at 371c, 5% C02, 100% humidity incubator. MTT assay and collagen synthesis were used as a method of this paper.

The obtained results were as follows

1. The cellular activity of gingival fibroblasts was significantly increased in 10-4g/ml of Carthamus tinctorius L. at the culture of the first, the second and the third day.
2. The cellular activity of periodontal ligament cells was significantly increased in 10-3g/ml of Carthamus tinctorius L. at the culture of the first, the second and the third day.
3. The cellular activity of osteoblasts was increased and maintained in 10-3g/ml and 10-4g/ml of Carthamus tinctorius L. but was significantly decreased over the level of 10-3g/ml.
4. The collagen synthesis of periodontal ligament cells was significantly more increased in 101¢¥g/ml than that of the control. and the collagen synthesis of osteoblasts was significantly increased in
10-3 g/ml too.
These findings suggest that increased cellular activity and collagen synthesis by Carthamus tinctorius L may help the periodontal regeneration and reconstruction.
